摘  要:目的观察大鼠骨关节炎(OA)模型中诱生型一氧化氮合酶(iNOS)与软骨退变严重程度相关性。方法建立前交叉韧带切断大鼠骨关节炎模型,按造模时间分为实验2、4、8周和假手术组、空白组,对各组软骨进行解剖显微镜下OA评分,iNOS免疫组织化学染色,比较iNOS在各组中的表达。结果iNOS的阳性细胞分数在空白组、假手术组、实验2、4、8周组分别为5.30±3.06、5.50±3.55、32.40±13.50.52.60±18.44、73.50±19.12,随造模时间延长而增加。iNOS的阳性细胞分数在OA评分为0、1、2、3时分别为5.00±4.55、45.20±9.50、60.50±14.30、79.50±16.20,随OA病变加重而增多。结论iNOS与软骨退变严重程度关系密切。Objective To observe the relationship between nitric oxide and the severity of cartilage degeneration in experimental ostcoarthritis ( OA ) model. Methods Rat OA model was established through transecting anterior crueiate ligment. Animals were divided into control, sham-operated, and experimental 2-,4-,8-week groups. Knees were harvested to observe the macro-pathologic changes of the cartilage, and score was evaluated. The iNOS expression was detected by immunohistochemistry. Results In control, sham-operated, and experimental 2-,4-, 8-week groups, the percentage of iNOS positive cells was 5.30 ± 3.06,5.50 ± 3.55,32.40 ± 13.50,52.60 ± 18.44 and 73.50 ± 19.12, respectively. The percentage of iNOS positive cells was 5.00 ±4.55,45.20 ±9.50,60.50 ± 14.30 ,and 79.50 ± 16.20 respectively when score was 0,1,2, and 3. With the severity of OA increased, the percentage of iNOS positive cells was increased. Conclusion iNOS is expressed in OA cartilage, and the iNOS expression has a close relationship with the severity of OA.
