摘 要:目的观察心复康口服液对压力负荷性心衰大鼠心肌细胞线粒体呼吸酶复合物活性的影响。方法将75只SD大鼠随机分为假手术组、腹主动脉缩窄组(模型组)、心复康口服液治疗组(治疗组)。采用腹主动脉部分缩窄法制作压力超负荷性大鼠模型。分别于药物干预10、12周时,应用分光光度法测定心肌细胞线粒体呼吸酶复合物Ⅰ-Ⅳ的活性变化。结果与假手术组相比,造模后第10周,模型组大鼠心肌细胞线粒体呼吸酶复合物Ⅱ(SDH)、Ⅳ(CCO)的活性显著性降低(P<0.01);和模型组相比,造模后第10周时治疗组大鼠心肌细胞线粒体呼吸酶复合物Ⅱ、Ⅳ的活性显著性升高(P<0.01),造模后第12周时上述改变更显著(P<0.01)。模型组大鼠12周时心肌细胞线粒体呼吸酶复合物Ⅱ(SDH)活性较10周时降低(P<0.05),治疗组大鼠呼吸酶复合物Ⅱ(SDH)、Ⅳ(CCO)活性在12周明显时高于10周(P<0.05)。结论心复康口服液能够明显提高CHF大鼠心肌细胞线粒体呼吸酶复合物的活性,改善心肌能量代谢以治疗心衰。Objective To observe the effect of "Xinfukang Oral Liquid" on the activities of myocardium mitochondrial respiratory complex enzymes( Ⅰ -Ⅳ) in pressure overload-induced congestive heart failure(CHF) rats. Methods Total 75 SD rats were randomly divided into three groups: sham operation (SH) group, model group and treatment group by "Xinfukang Oral Liquid". CHF model was established by partly constricting the abdominal aorta of rats. The activities of respiratory complex enzymes( Ⅰ -Ⅳ) were respectively detected by spectrophotometric method at the 10th and 12th weeks after treatment. Results Compared with the SH group, the activities of respiratory complex enzymes Ⅱ (SDH) and Ⅳ(CCO) were obviously decreased( P 〈0.01 ) at the 10th week; compared with model group, the activities of respiratory complex enzymes Ⅱ (SDH) and Ⅳ(CCO) in treatment group were increased evidently at the 10th week(P 〈0.01 ), and the increase was more obvious at the 12th week( P 〈0.01 ). The activity of SDH in model group at the 12th week was decreased compared with that of at the 10th week (P 〈0.05), and activities of SDH and CCO in treatment group were obviously higher at the 12th week than at the 10th week (P 〈0.05). Conclusion "Xinfukang Oral Liquid" can significantly improve activities of respiratory complex enzymes and myocardial energy metabolism to treat heart failure.
