摘 要:利用比较基因组学的方法从大白菜基因组序列中鉴定出一个新的LEAFY基因BrLFY1,并对它的序列特征、遗传进化和顺式元件进行了分析。结果表明,BrLFY1含有两个内含子,预测编码区为1260 bp,编码一个分子量为46.38 ku的蛋白。在进化上,BrLFY1与萝卜的RsLFY1关系最近,二者预测编码蛋白的序列一致性高达94.79%。另外,在BrLFY1的上游序列中存在多个激素和逆境应答元件。这说明大白菜LEAFY基因BrLFY1的表达可能不仅受多种激素的调节,还受多种逆境因子的影响。本研究为进一步研究BrLFY1在大白菜开花和逆境响应中的功能奠定了基础。Through the comparative genomics, a novel LEAFY gene BrLFY1 was identified from the genome of Chinese cabbage (Brassica pekinensis). Its sequence character, phylogenetic relationship with other plant' s LFYs and cis - elements were analyzed. The results showed that the genomic sequence of the gene contained 2 introns; its full length coding sequence was 1 260 bp, which encoded a putative 46. 38 ku protein. The phylogenetic analysis showed that BrLFY1 was the closest to Raphanus sativus gene RsLFY1, and their homology reached to 94. 79%. In addition, there were many putative cis - elements in the upstream of BrLFY1, which could be responsive to different kinds of hormones and environmental stresses. It was suggested that the expression of BrLFY1 might not only be regulated by hormones such as gibberellin, ethylene and auxin, but also by adverse environmental factors such as heat, drought and pathogens. This study laid a foundation for further functional study of BrLFY1 in flowering and response to environmental stress.
