摘　　要：目的:探讨N-乙酰基-丝氨酰-天冬氨酰-赖氨酰-脯氨酸(AcSDKP)对人真皮成纤维细胞增殖及胶原合成的影响。方法:人真皮成纤维细胞的原代培养及鉴定后,给予不同浓度AcSDKP(10-7,10-8,10-9,10-10,10-11mol/L)的干预,设立空白对照组。WST-8法检测人真皮成纤维细胞增殖;RT-PCR技术检测人真皮成纤维细胞Ⅰ、Ⅲ型胶原mRNA表达。结果:与空白对照组相比,10-10mol/LAcSDKP抑制人真皮成纤维细胞增殖作用最强;10-8mol/L和10-9mol/LAcSDKP对Ⅰ、Ⅲ型胶原mRNA表达的抑制作用最显著。结论:本实验发现AcSDKP能够抑制人真皮成纤维细胞增殖及Ⅰ、Ⅲ型胶原的合成,有望为病理性瘢痕的防治提供新方法。Objective To study the effects of N-acetyl-seryl-aspartyl-lysyl-praline （AcSDKP） on proliferation and synthesis of collagen in human dermar fibroblasts. Methods After primary cultures and identifications of human dermal fibroblasts, they were subjected to AcSDKP （10^-7, 10^-8, 10^-9, 10^-10, 10^-11mol/L, respectively）, and control group was established.The proliferation of human dermal fibroblast was detected by WST-8.The expression of COL Ⅰ and COL Ⅲ mRNA were analyzed with RT-PCR. Results Compared with control group,the proliferation of human dermal fibroblast was significantly inhibited by 10^-10mol/L AcSDKP.10^-8mol/L and 10^-9mol/L AcSDKP significantly inhibited the expression of COL Ⅰ mRNA and COL Ⅲ mRNA. Conclusion AcSDKP can significantly inhibit the proliferation and expression of COL Ⅰ ,COL Ⅰ of human dermal fibroblast. It maybe provide a new method to prevent and cure pathorogical scar.
