Establishment and evaluation of myocardial infarction model in rats

摘  要:目的初步探讨大鼠心肌梗死模型的建立及评价方法。方法采用冠状动脉左前降支大鼠结扎法建立心肌梗死模型,与10只正常大鼠进行对照。超声评价室壁厚度、左室收缩功能,病理学Masson's染色评价模型是否成功建立。结果40只大鼠成功建立模型29只,左室乳头肌短轴观显示建模大鼠左室前壁变薄,M型超声显示该处运动减弱、消失,左室短轴缩短率和左心室射血分数低于对照组(P<0.05)。Masson's染色显示左室前壁心肌梗死区呈蓝色染色。结论冠状动脉左前降支结扎法能成功建立大鼠心肌梗死模型,超声与Masson's染色能准确、有效评价心肌梗死模型是否成功建立。Objective To explore the establishment and evaluation methods of myocardial infarction (MI) model in rats. Methods MI model was established by coronary artery ligation, then the results were compared with those of 10 contrast rats. Their wall thickness and left ventrieular systolic function were evaluated by uhrasound, MI model establishment was evaluated by Masson' s staining. Results MI model was established successfully in 29 rats. Short axis view of left ventricular papillary muscle showed that the anterior wall of left ventricle became thinner, and M - mode ultrasound showed asynersis in that position. Left ventricular shortening fraction and left ventricular ejection fraction were lower than those in control group (P 〈0. 05 ). Masson' s staining showed that the MI area of left ventricle anterior wall was dyed into blue. Conclusion Left anterior descending coronary artery ligation can establish MI model in rats successfully, while ultrasound combined with Masson' s straining can evaluate MI model establishment accurately and effectively.
