摘 要:运用RT-PCR技术扩增鸭H-FABP基因序列,并用半定量RT-PCR方法研究该基因的组织特异表达规律。从35周龄母鸭心脏组织中首次克隆了H-FABP基因的cDNA序列,长度为606 bp,包含1个402 bp的完整编码序列,编码133个氨基酸。该序列与鸡H-FABP基因序列有约90.5%的同源性,而相应氨基酸序列的同源性为88.0%;与人、小鼠、牛、猪和山羊等哺乳动物的H-FABP基因序列的同源性在71.9%和76.6%之间,而相应氨基酸序列的同源性在72.9%和76.7%之间。半定量RT-PCR结果表明,该基因在心脏组织中的表达量最高,在骨骼肌、脂肪、小肠、腺胃、肌胃、肺、间脑和肾组织中均有中度或微量表达,而在肝、脾和卵巢中不表达。表明H-FABP基因在动物进化中具有高度保守性,具有相似的生物学功能,该基因的表达具有组织特异性。The sequence of duck H-FABP gene was obtained by reverse transcription PCR (RT-PCR) and analyzed, and its expression in different tissues was studied by semi-quantity RT-PCR. A 606 bp cDNA sequence of H-FABP gene was cloned from the heart of a 35-week-old female duck,which contained a complete 402 nucleotide coding sequence (CDS) ,encoding 133 amino acids. The duck H-FABP CDS shared 90. 5% homology with that of chicken, and about 71.9% -76.6% homology with that of mammal (such as human, mouse, cattle and pig) ; while the corresponding protein shared 88.0% and 72. 9% -76. 7% ,respectively. The results of the semi-quantity RT-PCR indicated that duck H-FABP mRNA has the highest expression level in heart, and middle or low expression in skeletal muscle, adipose, small intestine, glandular stomach,muscular stomach, lung, diencephalons and kidney, and did not expressed in liver, spleen and ovary. H-FABP gene is of high conservation in the course of animal evolution, and they have the approximate biological function in different animals. The expression level of H-FABP mRNA is particular to tissues.
