摘 要:目的建立血浆高丰度蛋白去除方法,评价其应用价值。方法应用Bio-rad公司生产的血清白蛋白和IgG去除试剂盒对血浆样品进行处理,比较去除和未去除高丰度蛋白血浆2-DE图谱。结果血浆高丰度蛋白经去除后,血浆样品中的低丰度蛋白得到较好的分离和浓缩。全血浆2-DE图谱分离的蛋白质点176个,而去除高丰度蛋白后血浆2-DE图谱上分辨率增高,分离出蛋白质点287个,一些低丰度蛋白质被有效分离出来。结论Bio—rad公司生产的血清白蛋白和IgG去除试剂盒可有效提高低丰度蛋白的检出敏感性,为血浆蛋白质组学研究提供了很好的工具。Objective to establish the method for removing high- abundance proteins in plasma proteomics research and to estimate the value of the method. Methods High-abundance proteins were removed from plasma sample by using ProteoPrep Immunoaffinity Albumin and IgG Depletion Kit of Bio-rad company. The residues were determined by two-dimensional polyacrylamide gel electrophoresis (2-DE). Results The low-abundance proteins in plasma was highly enriched after the removal of high- abundance proteins. 176 protein dots were observed in the intact plasma whereas 287 protein dots were effectively separated when high-abundance proteins were removed. Some low-abundance proteins were also visualized after this process. Conclusions Detecting sensitivity of low-abundance proteins can be effectively enhanced by ProteoPrep Immunoaffinity Albumin and IgG Depletion Kit of Bio-rad company.
