摘 要:目的:筛选出对多种肠道病毒具有抗病毒效力的SiRNA序列。方法:比对多种肠道病毒5'端非编码区(NCR),针对其共有序列设计3段SiRNA,分别转染入感染肠道病毒CoxB3,Echo6,和Enterovirus 71的PC12细胞。RT-PCR检测细胞病毒基因表达情况,观察细胞形态变化,MTT检测细胞生存率。结果:SiRNA序列成功转染入PC12细胞。序列1、序列3SiRNA对CoxB3病毒、Echo6病毒及Enterovirus 71病毒复制具有抑制作用,序列2SiRNA对CoxB3病毒及Echo6病毒具有抑制作用。结论:本实验选择针对肠道病毒5'-NCR区域的共有序列设计的SiRNA能抑制多种常见肠道病毒复制,有望为临床治疗肠道病毒性脑炎、心肌炎提供新的资料。Objective:To find out several effective SiRNA sequences against enteroviruses. Methods:Several SiR-NA sequences aiming at the consensus sequence of non-coding regions of enteroviruses were synthesized, and transfected into PC12 cells infected with 3 kinds of enteroviruses (CoxB3, Echo6, and Enterovirus71) respectively. RT-PCR method was used to detect the gene expression of enteroviruses in PC12 ceils, and MTT to measure the survival rate. Cell morphological changes were observed microscopically. Results: SiRNA sequences were transfected into PC12 cells successfully. The 1st and 3rd SiRNA sequences could restrain viral replication of CoxB3 virus, Echo6 virus and Enterovirus71 in PC12 cells, and the 2nd SiRNA sequence could suppress CoxB3 virus and Echo6 virus in PC12 cells. Conclusion:The three SiRNA sequences which were designed for the consensus sequence in 5′-NCR of enteroviruses can restrain replication of many kinds of enteroviruses.
