摘  要:目的:探讨土荆皮酸(PAB)和顺铂联用对人卵巢癌细胞株HO-8910裸鼠皮下移植瘤的生长及其血管生成的影响。方法:将20只荷人HO-8910卵巢癌的裸小鼠随机分成4组:①PAB组(60 mg/kg,腹腔注射治疗5天);②顺铂组(5 mg/kg,腹腔注射治疗5天);③联合用药组(PAB 60 mg/kg+顺铂5 mg/kg,腹腔注射治疗5天);④阴性对照组(给予同体积的含有6%聚乙二醇、3%乙醇、1%Tween 80的水溶液);计算瘤质量、瘤体积;同时应用CD34免疫组化染色,观测瘤组织内的微血管密度。结果:统计学分析表明:在抑瘤率和微血管密度方面,PAB组、顺铂组和阴性对照组比较差异具有统计学意义(P<0.01),联合用药组和其他3组比较差异均有统计学意义(P<0.01)。结论:PAB、顺铂对人卵巢癌移植瘤的生长具有抑制作用;PAB和顺铂联用能起增效作用。Objective:To explore the effect of pseudolaric acid B(PAB)and cisplatin on growth and angiogenesis of xenografted tumor of human ovarian cell line HO-8910 in nude mice.Methods:20 nude mice bearing human ovarian carcinoma HO-8910 xenografted tumor were divided into 4 groups randomly: ①PAB group(injected by 60 mg/kg PAB for 5 days);②cisplatin group(injected by 5 mg/kg cisplatin for 5 days);③combination group(injected by 60 mg/kg PAB and 5 mg/kg cisplatin for 5 days);④control group(same volume of solution containing 6% polyethylene glycol,3% ethanol and 1% Tween 80);the tumor volume and weight were measured;the microvessel densities(MVDs) in four groups were detected by CD34 immunohistochemical staining.Results:There was difference in inhibition rate of tumors and MVD between PAB group,cisplatin group and control group(P〈0.01) as well as between combination group and the other 3 groups(P〈0.01).Conclusion:PAB and cisplatin can inhibit tumor growth of nude mice bearing human ovarian carcinoma cell line HO-8910 and their combination can produce a synergistic action.
