摘  要:目的探索脐血采集后单个核细胞(MNC)最佳分离和冷冻的时间窗。方法采集脐血6份,每份按采集后分离时间分为8h组、24h组、48h组和72h组。每组标本均在分离后进行MNC计数及台盼蓝染色检测细胞活率;各时间组细胞活率在50%以上的行体外CFU形成能力检测及玻璃化冷冻。冷冻10h后解冻进行细胞活率及体外CFU形成能力检测。结果冷冻72h组细胞活率低于其他组(P<0.05);解冻后72h组MNC计数低于24h和48h组(P<0.05);48 h以后分离的MNC解冻后细胞活率降低(P<0.05)。结论脐血MNC采集后的48h内分离冷冻不影响细胞活率及冻存后集落形成能力。Objective To demonstrate a time window for separation and cryo -preservation of cord blood after being collected. Methods After being collected and separated in 8h, 24h, 48h, 72h, 96h, the cord blood MNC were counted and tested its viability by typan blue stain. The cells which viability was over 50% were cryopreserved by vitrification cryopreserved method and the recovered cells were tested its viability by typan blue stain. Recover rate was counted by MNC and CFU-GM. Results The viability of the cells which separated in 72h was significantly lower than that of the other group (P〈0.05) ; After cryopreserving, the MNC counting and viability of 72h group were lower significantly than those of the other group (P〈0.05) ; There were no significant difference in viability, recover rate and the receiver rate of CFU-GM within 48h after collecting. Conclusion The right time to separate the cord blood is within 48h after collecting, but the best time is within 24h after collecting.
