摘 要:本研究采用AFLP技术,对2005年引自加拿大的海扇贝(Placopecten magellanicus)以及中国北方主要养殖的虾夷扇贝(Patinopecten yessoensis)、栉孔扇贝(Chlamys farreri)和海湾扇贝(Argopecten irradians)进行了遗传学比较分析。结果表明,共用5对AFLP引物进行了扩增,共扩增出501个位点,多态性位点为392个,多态位点比例为78.24%。平均Nei氏基因多样性指数H分别为0.2139、0.1684、0.1441和0.1670,平均Shannon氏指数I分别为0.3356、0.2681、0.2439和0.2656。海扇贝多态位点比例、Nei氏基因多样性指数和Shannon氏指数均处在较高水平,说明海扇贝遗传多样性较高。UPGMA聚类结果表明,海扇贝与虾夷扇贝亲缘关系最近。AFLP technology was used to analyze the genetic diversity of Placopecten magellanicus which was introduced into China in 2005, and other three main cultured scallop species including Patinopecten yessoensis, Chlamys farreri and Argopecten irradians. Five pairs of primers were used and 501 loci were detected, 392 of them were polymorphic, percent of polymorphic loci were 78.24%. Nei's gene diversity index were 0.2139, 0.1684, 0.1441 and 0.1670, while Shannon indexes were 0.3356, 0.2681, 0.2439 and 0.2656, respectively. The results showed that Placopecten magellanicus had higher level of polymorphic loci, Nei's gene diversity index and Shannon index, that was the higher level of genetic diversity than other three cultured scallop species in China. UPGMA results showed that P. magellanicus and P. yessoensis had the closest genetic relationship than others. This research would be a first try on genetic analysis in P. magellanicus after its introduction, also would be a basis on genetic breeding and the creation for new varieties of scallop species in China.
