摘 要:目的建立测定水飞蓟宾纳米结构脂质载体中主药含量及包封率的方法。方法采用高效液相色谱法测定药物含量,色谱柱为Kromasil C18柱(4.6mm×150mm,5μm),流动相为甲醇-0.35mol·L-1醋酸(48:52),流速为1mL·min-1,检测波长为288nm;分别采用离心超滤法、葡聚糖凝胶过滤法分离水飞蓟宾纳米结构脂质载体和游离药物以测定包封率。结果水飞蓟宾在1~40mg·L-1内线性良好(r=0.9998),平均回收率为99.57%(RSD=0.52%),离心超滤法及葡聚糖凝胶过滤法测得平均包封率分别为96.87%,84.20%。结论采用离心超滤法分离脂质载体与游离药物,高效液相色谱法测定药物含量,可高效、准确、方便地测定水飞蓟宾纳米结构脂质载体包封率。OBJECTIVE To establish a RP-HPLC method for the determination of the content and entrapment efficiency of silybin nano-structured lipid carrier (NLC). METHODS The content of silibin NLC was determined by HPLC.The analytical column was Kromasil C18 (4.6 mm× 150 mm,5μm) and the column temperature was ambient. The mobile phase was methanol-0.35 mol·L^-1(48 : 52). The flow rate was 1 mL·min^-1. The detection wavelength was 288 nm. Centrifugation ultrafiltration and sephadex gel filtration methods were employed to isolate free silybin for the entrapment efficiency. RESULTS Silybin was linear in the range of 1-40 mg·L^-1 (r=0.999 8), the average recovery was 99.57%(RSD=0.52%). The entrapment efficiencies of silybin NLC determinated by sephadex gel filtration and centrifugation ultrafiltration were 84.20% and 96.87%, respectively. CONCLUSION Centrifugation ultrafiltration combined with HPLC is able to determine the entrapment efficiency of silybin NLC efficiently, accurately and conveniently.
