摘　　要：爱维莫潘是一种新型的口服外周阿片样受体拮抗剂,用于治疗术后肠梗阻(POI)。该药对μ阿片样受体亲和性比对κ和δ阿片样受体高。本品仅对外周受体有作用,其分子极性限制胃肠道吸收,不能进入血脑屏障。Ⅲ期临床研究证明爱维莫潘耐受性较好,能够有效加快腹部手术后胃肠功能恢复,各剂量组的不良反应与安慰剂组相似。现就爱维莫潘的药理作用、药动学、药物相互作用及安全性评价进行综述。Alvimopan, a novel and peripherally acting μ-opioid receptor antagonist, is currently under evaluation for the management of postoperative ileus （ POI）. Alvimopan has a greater affinity for the μ-receptor than for the κ- or δ-opioid receptors. The polarity of its molecule limits gastrointestinal absorption and central nervous system penetration, so only has peripheral activity. Completed phase Ⅲ trials suggest the tolerability and efficacy in accelerating the recovery of gastrointestinal function after abdominal surgery. Adverse events caused by all doses are similar to those in placebo groups. In this paper, we reviewed the pharmacology, pharmacokinetics, drug interactions and safety of alvimopan.
