摘 要:目的探讨临床不同分离株阴道毛滴虫核糖体DNA基因转录间隔区(rDNA ITS)序列多态性与甲硝唑抗性表型之间的相关性。方法采用微孔板法检测39株达到纯培养的阴道毛滴虫虫株对甲硝唑敏感性,同时设计rDNA-ITS区特异引物进行PCR扩增并测序。结果39个虫株中,甲硝唑敏感虫株20株,耐药虫株19株;PCR扩增获得ITS区和5.8 S特异片段长度为285 bp。经DNAStarM egA lign比对显示,仅在ITS1区存在唯一点突变,即C66T突变。39个虫株中有8株具C66T突变,其中6株来自甲硝唑敏感虫株,2株来自甲硝唑耐药虫株。经F isher确切概率检验,χ2=0.108,P>0.05。结论阴道毛滴虫临床分离株ITS区C66T突变与甲硝唑抗性表型无明显相关性。Objective To study the relationship between the polymorpholism of rDNA-ITS and the resistance to metronidazole in Trichomonas vaginalis. Methods Thirty-nine clinical isolated strains of Trichomonas vaginalis were detected for resistance to metronidazole in aerobic condition with 96-wells plates and rDNA-ITS were amplified and sequenced. Results Twenty strains were metronidazole susceptible and 19 strains were metronidazole resistant. A 285bp rDNA-/TS and 5.8S fragment were amplified and sequenced. Only one point mutation (C66T) was detected by sequence alignment with MegAlign of DNAStar software. A total of 8 strains were detected in which 6 strains were from metronidazole-susceptible group and the other 2 from the metronidazole resistant group. There was no statistics difference between susceptible and resistant strains with Fisher exact test ( P 〉 0.05, X^2 = 0. 108). Conclusion There is no obvious association between C66T mutation of rDNA-ITS and resistance to metronidazole in T. vaginalis.
