摘  要:目的观察复方清热颗粒剂对耐药大肠杆菌外膜通透性的干预作用。方法大肠杆菌经各药物组作用后,提取外膜蛋白,行蛋白电泳和紫外凝胶成像,以外膜蛋白相对含量为指标,观察中药复方清热颗粒原液、含药血清、中西药结合作用前后耐药大肠杆菌外膜通透性的变化。结果舒普深+含药血清作用后,能使4株大肠杆菌外膜蛋白OmpF含量增加;舒普深+中药原液作用后,能使大肠杆菌ATCC35218(产β-内酰胺酶株)、大肠杆菌ATCC25922(标准敏感质控株)、临床产ESBL大肠杆菌ESBL1的外膜蛋白OmpF含量增加;中药原液能使大肠杆菌ATCC25922(标准敏感质控株)外膜蛋白OmpF含量增加;单纯含药血清作用后4株大肠杆菌的外膜OmpF含量无增加。结论中西药结合能降低耐药大肠杆菌的耐药性,可能是通过增加大肠杆菌外膜通透性而发挥作用。Objective: To observe the action of the Compound Qingre Granules on the outer membrane permeability of the drug - resistance E. coli. Methods: After being affected by several drugs, the outer membrane proteins of E. coli were extracted. Then the electrophoretic analysis and the ultraviolet gel imaging system analysis were carried out. Moreover, the granules original solution, the drug serum and the variations of the outer membrane permeability of the drug resistance E. coli were investigated by using out membrane proteins. Results: After using the sulperazon and the drug serum, the concentrations of OmpF of the four strains of E. coli have increased. After using of sulperazon and granules original solution, the concentration of OmpF of the E. coli ATCC 35218, ATCC25922, clinical producing ESBL E. coli and out membrane proteins of ESBL1 have increased. However, in pure drug serum, the concentrations of OmpF of the four strains E. colis remain unchanged. Conclusion: The collaborative treatment could decrease the drug resistance of E. col/ by increasing of the outer membrane permeability of E. coli.
