摘  要:测定大环柄菇属一种真菌(Macrolepiotasp.)的rDNA ITS序列,根据序列构建系统发育树;采用固体培养方法在不同培养基和不同培养条件下对该菌菌丝体的生长条件进行分析,最后采用正交试验,考察碳源、氮源、pH值和矿质元素对该菌菌丝生长的影响。结果表明,Macrolepiotasp.与来自日本的Macrolepiota sp.Oku X-2000和Macrolepiota sp.TMI-23928亲缘关系近;正交试验结果表明,碳源、pH值、矿质元素和氮源对该菌菌丝体的生长速度有显著影响,固体培养条件优化结果为葡萄糖、豆饼粉、pH5且培养基中缺乏KCl。经优化后,菌丝体的生长速度可以达到2.28 mm.d-1。In this paper, the ITS region of nuclear ribosomal DNA of Macrolepiota sp. was sequenced, and a phylogenetic tree was set up based on ITS data. The solid culture method was selected to study the growth state of Macrolepiota sp. in different media and culture conditions. The results showed that the phylogenetic relationships of Macrolepiota sp. were closely related to Macrolepiota sp. Oku X-2000 and Macrolepiota sp. TMI-23928. The results of orthogonal experiment showed that C source, pH value, mineral element and N source had significant effect on the growth rate of the myeelia. The optimum solid culture conditions were glucose, bean cake powder, pH and lacking KC1 in the medium. The growth rate of mycelia was 2.28 mm· d^-1 under the optimized cultivation.
