摘 要:目的研究树突状细胞(DC)在慢性阻塞性肺疾病(COPD)大鼠模型肺部的表达状况,探讨吸入糖皮质激素布地奈德及抗胆碱能受体支气管扩张剂异丙托溴铵对COPD大鼠模型肺部DC的影响。方法用两次气道内滴入脂多糖(200μg/次)及熏烟4周的方法制作COPD模型(模型组)。药物干预组自第8d起分别吸入布地奈德及异丙托溴铵溶液。观察肺部病理改变,同时通过免疫组化的方法检测肺部DC分布及数量。结果模型组及药物干预组大鼠呈现COPD的特征性病理改变。模型组及药物干预组DC数目较对照组明显增高(P<0.01),布地奈德组较模型组DC数目减少(P<0.01),异丙托溴铵药物治疗组与模型组比较DC数目无统计学差异。结论DC细胞在COPD大鼠肺部表达明显增加,布地奈德干预可以使这种增加减少。Objective To evaluate the expression of dentritic cell (DC)in small airways of C OPD model rats,to observe the effects of inhaled budesonide and ipratropium bromide on the COPD model. Methods The rat COPD model (model group)was established by intratracheal instillation of lipopolysaccharide (LPS)twice and exposure to eigarate smoke. Drug inhalation group rats received inhalation of Pulmicort and Atrovently since the second week. The pathologic characteristics of bronchial walls were determind- ed. At the same time the numbers and the distribution of DCs in each group were investigated by immunohistoehemistry. Results COPD model rats showed .typical pathological and pathophysiological features. There is a significant increase in DC number in lungs of COPD model group rats compared with the control group rats(P〈0.01 ). DC number in budesonide-treatment group also in- creases,but less than that in model group (P〈0.01). There are no difference between the ipratropium bromide treatment group with the model group(P〉0.05). Conclusions There is significant increase in DC number in lungs of COPD model group than control group. Giving budesonide-treatment will reduce this change.
