摘 要:目的本试验旨在研究β-胡萝卜素-15,15’-加单氧酶(βCMOOX)的组织特异性。方法使用半定量RT-PCR的方法研究三黄鸡体内βCMOOXmRNA在不同组织内的表达,并用高效液相色谱法测定各组织中βCMOOX的活性。结果研究表明βCMOOXmRNA在正常鸡的心脏、肝脏、脾脏、肾脏、肠道、睾丸和骨骼肌组织中均有表达,且在空肠内表达水平最高(P<0.01),在肺中几乎没有表达;βCMOOX在各组织内(除肺脏外)均有活性,且空肠和十二指肠内活性最高(P<0.05)。结论β-胡萝卜素15,15’-加单氧酶在鸡体内的分布具有组织特异性。Objective To study the tissue-specificity of β-carotene-15,15'-monooxygenase( β CMOOX). Method Semi-quantitative RT-PCR and HPLC were used to study the expression of βCMOOX mRNA and its activity in different tissues of chicken. Results βCMOOX mRNA was expressed in tissues including heart, liver, spleen, lung, kidney, duodenum, jejunum, ileum, muscle and testis, not expressed in lung. Furthermore, it was expressed in jejunum with the highest level. βCMOOX activity was found in every tissue, except lung, and highest in duodenum and jejunum. Conclusion The distribution of βCMOOX in chicken has tissue specificity.
