摘  要:目的研究不同处理时间和不同剂量的维生素C(VC)对神经元细胞生长的影响。方法体外原代培养的新生SD乳鼠大脑皮层神经元细胞,在细胞生活的不同时间(D1、D4、D7、D14)加入不同浓度的VC(0~800μmol/L)共培养,用MTT法检测细胞存活率,并选取D7加入VC共培养的细胞作后续研究,观察VC对神经元细胞的突起数目、长度、胞体面积和长轴长度的影响。结果MTT结果显示:D1加入VC,800μmol/L处理组使细胞存活率降低;D4或D14加入VC,400μmol/L处理组使细胞存活率提高,而800μmol/L处理组使细胞存活率降低;D7加入VC,200、400μmol/L组的MTT值明显高于对照组,800μmol/L组的MTT值低于对照组。进一步观察发现,在D7加入VC,无论浓度如何,对细胞突起的数目无明显影响;而200、400、800μmol/L处理组能使突起长度、细胞胞体面积和长轴长度较对照组显著增高,作两两比较,400μmol/L组高于其他组。结论在细胞生长的D7加入400μmol/L的VC,其对神经元细胞的生长有明显的促进作用。Objective To investigate the dose-time effect of vitamin C treatment on the growth of cortical neurons. Method On D1, D4, D7 or D14 d of cultured cortical neurons from the newborn SD rat, different concentrations of VC (0-800 μmol/L) were added to the culture medium. The livability was measured by MTT method. The cells which medium was added VC on D7 were used for research in the study. The effects of VC on the number and length of neuronsal processes as well as the area and maximal diameter of neurons were observed. Results In the cells treated by VC on D1, the livability of 800 μmol/L group was significantly lower than that of control group. In the cells treated by VC on D4 or D14, the livability of 400 μmol/L group was significantly higher than that of control group, while that of 800 μmol/L group was lower too. In the cells treated by VC on D7, the livability of 200 μmol/L and 400 μmol/L groups was significantly higher than that of control group, while that of 800 μmol/L group was lower too. In the cells treated by VC on D7, the number of neuronal processes were not effected from 0- 800 μmol/L VC. But the length of neuronal processes as well as the area and maximal diameter of neurons were higher in 200, 400, and 800 μmol/L groups than those in control group, the highest in 400 μmol/L group. Conclusion 400 μmol/L VC could improve neuron growth on D7 of cultured neurons in vitro.
