摘 要:目的:检测斑秃患者外周血单核细胞STAT4和血清IL-12及IFN-γ的表达。方法:应用RT-PCR方法检测50例斑秃患者和25名正常人外周血单核细胞(PBMC)中STAT4的mRNA表达;ELISA法检测IL-12和IFN-γ的表达。结果:活动期斑秃患者中IL-12、STAT4和IFN-γ的水平显著高于缓解期患者(均P<0.05)和正常人(均P<0.05);病程>3个月的患者IL-12、STAT4和IFN-γ的表达显著高于病程<3个月的患者,但二者均高于正常人。结论:斑秃患者中存在IL-12和STAT4的高表达,可能通过IL-12/STAT4/IFN-γ途径直接或间接诱导IFN-γ的过量表达,导致毛囊部位炎症形成,引起脱发。Objective: To detect the expression of IL- 12, STAT4 and IFN -γ. Methods: Real time PCR was applied to detect the expression of STAT4 mRNA in peripheral blood mononuclear cell (PBMC), and ELISA was used to determine the level of IL - 12 and IFN -γ in sera from 50 patients with alopecia areata and 25 healthy controis. Results: The expression of IL - 12, STAT4 and IFN -γ was significantly higher in active alopeeia areata patients than in inactive alopeeia areata patients ( P 〈 0.05 ) and normal controls ( P 〈 0.05). The expression of IL - 12, STAT4 and IFN - γ was significandy higher in the patients with a duration more than 3 months than the patients with a duration less than 3 months ( P 〈 0.05). Conclusion: The high expression of IL- 12 and STAT4 in alopecia areata may induce inflammation of hair follicles and alopecia caused by over - production of IFN - γ, which is induced through a IL- 12/STAT4/IFN-γ/activation pathway directly or indirectly.
