摘 要:目的观察NF-κB对原发性肾病综合征患儿IL-8调控及黄芪干预作用。方法在原发性肾病综合征患儿和正常儿童外周血单个核细胞中,分别加NF-κB的刺激剂、抑制剂及黄芪,测IL-8浓度和NF-κB活性。结果肾病患儿NF-κB活性及IL-8含量增高,2者呈正相关;IL-1β刺激后,黄芪降低肾病患儿NF-κB活性及IL-8含量、降低正常儿童NF-κB活性而不影响IL-8含量;用对甲苯磺酰-L-苯丙氨酸氯甲基甲酮(TPCK)抑制后,黄芪可提高肾病患儿NF-κB活性及IL-8含量、提高正常儿童NF-κB活性;而不影响IL-8含量。结论原发性肾病综合征患儿NF-κB活性上调及IL-8水平增高,黄芪通过抑制NF-κB过度活化从而下调IL-8过高表达继而发挥效应;黄芪对NF-κB活性有双向调节作用。Objective To study the adjustments of IL-8 caused by NF-κB in the patients with primary nephritic syndrome(PNS) and the intervention of Astragalus. Methods NF-κB stimulator,NF-κB inhabitor and Astragalus were added to the different tubes of peripheral blood mononuclear cells(PBMC) respectively.The concentrations of IL-8 and the activity of NF-κB were detected. Results The activity of NF-κB and the level of IL-8 in PNS children are higher than those of normal children. In the PNS children ,these two factors had a positive linear correlation. The Astragalus can decrease the activity of NF-κB and the concentration of IL-8 in PNS patients. PBMC treated with stimulator IL-1β, Astragalus can decrease the activity of NF-κB and the concentration of IL-8 in patients with PNS. It also decreases the the activity of NF-κB in normal children , but can' t affected the concentration of IL-8 in normal children. PBMC treated with inhibitor N- tosyl phenylalanine chlormethyl ketone(TPCK) , Astragalus can increase the activity of NF-κB and the concentration of IL-8 in patients with PNS. it also increases the the activity of NF-κB in normal children ,but can't affected the concentration of IL-8 in normal children. Conclusion In patients with PNS, the activity of NF-κB was up-regulated and the level of IL-8 was significantly elevated. Astragalus can take effect by inhibiting over-active NF-κB to downregulated the over-expressed IL-8 in patients with PNS. The dualdirectional regulation effects of Astragalus on signaling pathway of NF-κB was indicated.
