青海石鸡血清淀粉酶同工酶酶谱及其活性研究

摘  要:[目的]为青海石鸡的生化遗传学研究提供基础资料。[方法]采用聚丙烯酰胺凝胶垂直平板电泳法对39只青海石鸡血清淀粉酶同工酶酶谱进行研究,并利用比色法对其淀粉酶活性进行测定。[结果]青海石鸡血清淀粉酶有Amy-1,Amy-3 2种同工酶,其中Amy-1有AA、AB和BB 3种表型,表型频率分别为17.95%、66.67%、15.38%;AB为优势表型,Amy-1A和amy-1B等位基因频率分别为0.512 8和0.487 2,基因杂合度为0.499 7;青海石鸡血清淀粉酶活性为(566.62±112.20)U/L。[结论]青海石鸡Amy-1同工酶呈现多态性,Amy-1 AB表型的血清淀粉酶活性显著高于Amy-1 AA和Amy-1 BB表型。[ ObjeCtive] The aim of the study was to provide basic date for biochemical genetic study of Qinghai Alectoris graecu. [ Method ] 39 Qinghai Alectoris graeca were used to study isozyme pattern of serum amylase and activity with vertical polypropylene gel electrophoresis and spectrophotomettry, respectively. [ Result ] The study showed that there were two isozymes of serum amylase including Amy-1 and Amy-3 in Qinghai Alectoris graeca. There were three kinds of types that were AA,AB and BB in Amy-1 isozyme ,and the phenotype frequency of AA,AB and BB were 17.95 % ,66.67%, 15.38 %. The AB was the advantageous type, and the allele frequency of Amy-1 A and Amy-1B were 0.5128 and 0. 4872 respectively. The gene heterozygosity was 0.499 7. The average activity of serum amylase was (566.62 ± 112.20)U/L. [ Conclusion ] The Amy-1 isozyme existed the polymorphism in Qinghai Alectoris graeca. The activity of serum amylase in the Qinghai Alectoris graeca with Amy-1 AB phenotype was significantly higher than that in the Alectoris graeca with Amy-1 AA and Amy-1 BB phenotype.
