摘  要:目的研究不同培养基、不同植物激素和活性炭对天山雪莲组培苗生根的影响。方法采用7种培养基,3种植物激素和不同添加量的活性炭,通过测定天山雪莲的根长、根数和生根率3个生根指标来筛选最适生根培养基。结果有4种培养基可以促进生根,其中White培养基的效果最佳,根长达3.431cm,根数2条,生根率达80%;添加生长素IAA(吲哚乙酸)、IBA(吲哚丁酸)和NAA(萘乙酸)均可以促进生根,其中0.2mg·L-1的IAA的效果最佳,根长达到1.294cm,平均根数达到8条,生根率为100%;添加活性炭可以提高生根质量,最适添加量为1g·L-1。此时,平均根长为1.620cm,平均根数为9条,生根效果最佳。结论天山雪莲组培苗最适生根培养基为White+0.2mg·L-1IAA+1g·L-1活性炭。OBJECTIVE To study the effect of the growth medium, plant hormones and active carbon on rooting of in vitro shoot of Saussurea involucrata. METHODS Seven growth medium, three plant Hormones and different concentration active carbon were applied. Three rooting indexes, including rooting length, rooting number and rooting rate, were used to screen the optimal rooting medium. RESULTS Four growth mediums enhanced the induction of the rooting. The white medium produced the best result. The average root length was 3.431 cm, the average root number was 2, the rooting rate is 80%. The auxins enhance the induction of the rooting, such as IAA (indole-acetic acid), IBA(indole-3-butyric acid) and NAA(naphthalene-acetic acid). 0.2 mg.LI IAA showed optimum result considering the root length, the root number and the rooting rate. Active carbon improved the quality of the rooting. The optimal concentration was 1 g.L-1, the root length was 1.620 cm, the rooting number was 9. CONCLUSION The optimal rooting medium of Saussurea involucrata is White+0.2 mg·L-1 IAA+ 1 g·L-1 active carbon.
