摘 要:目的:建立一种简便、可靠、快速的肿瘤药敏试验新方法。方法:采用组织块培养、MTT染色和计算机图像分析法测定药物对肿瘤细胞的作用;测定加MTT后不同时间显色反应及非特异性染色以确定试验条件;对同一份标本进行3次试验以观察重复性;对人癌裸鼠移植瘤进行药敏试验以评价本试验鉴别敏感/耐药的能力;对127份临床肿瘤标本进行药敏试验,观察试验成功率。结果:加MTT反应时间以3h为合适,试验变异系数为9.4%,本试验可鉴别移植瘤的耐药性,对临床肿瘤标本试验成功率90.6%。结论:组织块培养、MTT染色和计算机图像分析的肿瘤药敏系统具有简便、可靠、快速的特点,可指导临床个体化肿瘤化疗。Objective :This study is to establish a simple operation, reliable and rapid manner for tumor chemosensitivity assay in vitro. Methods:The effect of drug on tumor cell was evaluated with tissue cuhure-MTT endpoint-image analysis system. The assay condition was determined by analyzing the stability of color reaction and the non-special color reaction after adding MTT. The repeat character was evaluated by 3 assays on same sample. The ability of the assay to distinguish drug-resistance was evaluated by the assays on cancer xenografts in nude mice of 4 pair human tumor sensitivity/drug-resistance cell lines. The success rate was evaluated by assays of 127 clinic tumor samples. Results :The optimum time of color reaction was at 3 hour point after adding MTT. The coefficient of variation was 9.4%. The assay distinguished drug-resistance. The success rate of assay of 127 clinic tumor samples was 90. 6% . Conclusion:The assay may be promising to develop as a useful method for individual cancer chemotherapy.
