摘  要:目的建立一种敏感、特异、快速诊断女性生殖道沙眼衣原体(CT)感染的方法。方法应用套式聚合酶链反应(NPCR)技术和直接荧光(DFA)法检测女性生殖道沙眼衣原体。对两者结果不符的标本用细胞培养法验证。结果300份受检标本,DFA法阳性71份,阴性229份,NPCR法阳性90份,阴性210份;其中NPCR和DFA均为阳性者68份,另有22份NPCR阳性,而DFA为阴性,再经细胞培养证实系阳性。但另有3份DFA阳性,NPCR却为阴性,再经细胞培养证实亦属阳性。综合NPCR与DFA并对照细胞培养,结果显示NPCR的敏感性为967%(90/93),特异性为100%(207/207),DFA的敏感性为763%(71/93),特异性为100%(207/207)。结论NPCR技术与DFA法检测CT感染,均具有高度特异性、操作简便等优势;NPCR的敏感性优于DFA法;DFA法具有费用低、重复好的特点。两法结合起来检测CT,将获得理想的检测效果。Objective To establish a method for sensitive, selective and speed diagnosis of chlamydia trachomatis (CT) infected in female genital tract. Methods CT in female genital tract was detected by NPCR and DFA. Discrepant results between NPCR and DFA were further analyzed by cell culture. Results In 300 tested samples, 68 were both positive reaction of NPCR and DFA, 22 were NPCR positive but DFA negative, whereas 3 were DFA positive and NPCR negative. These discrepant samples were further determined by cell culture and all were positive. NPCR had a sensitivity of 96% (90/93) and a specificity of 100%(207/207). DFA showed a sensitivity of 76.3%(71/93) and a specificity of 100%(207/207). Conclusions The results suggested that NPCR had a higher sensitive, selective and simple procedure, DFA is rapid, simple, sensitive and selective. Combined NPCR with DFA to detect CT, will show satisfied results.
