摘 要:观察二甲双胍对STZ糖尿病大鼠血糖和骨骼肌中葡萄糖转运体Ⅳ(GLUT4)基因表达的影响。方法STZ糖尿病大鼠每公斤体重100mg/d二甲双胍灌胃治疗3周后,斑点杂交测定骨骼肌中的GLUT4mRNA含量。结果糖尿病治疗组的血糖降低30.3%,较非治疗组低28.6%,而骨骼肌中GLUT4mRNA含量较非治疗组高65.6%。结论二甲双胍促进骨骼肌中GLUT4基因表达可能是其降低STZ糖尿病大鼠血糖的机制之一。PURPOSE To investigate the effect of metformin on glucose homeostasis and expression of glucose transporter Ⅳ(GLUT4) in the skeletal muscle of the streptozotocin induced diabetic rats. METHODS The STZ-diabetic rats were administered metformin by gavage (100mg/kg body weight/day) for 3 weeks. The GLUT4 mRNA in the skeletal muscles was assessed with dot blot analysis. RESULTS After 3 week treatment, the blood glucose in the metformin treated diabetic rats decreased by 30.3% , and was 28.6% lower than that of the untreated diabetic rats. Dot blot analysis showed that the GLUT4 mRNA in the skeletal muscle of treated diabetic rats was 65.6% higher than that of the untreated diabetic rats. CONCLUSIONS The hypoglycaemic effect of metformin in the STZ-diabetic rats may be due to the increased expression of the GLUT4 gene in the skeletal muscle.
