摘  要:研制一种可以选择性测定水溶液及生物样品中NO的较灵敏的新型修饰电极。方法在100μm铂电极表面依次用脱乙酰甲壳素、NiSO4和Nafion修饰,制成NO电流传感器,研究了NO在该电极上的电化学反应,并用于安培法测定大鼠海马脑片中NO的释放。结果NO可以在修饰电极表面催化氧化,氧化电流与NO浓度在4.5×10-4~6.7×10-8mol/L之间呈现良好的线性关系,检测下限为3.0×10-8mol/L,电极对抗坏血酸、多巴胺及亚硝酸根有较好的选择性。结论该电极的机械强度、灵敏度及选择性较高,适于NO的离体及在体测定。PURPOSE To prepare a new kind of modified electrodes that could be used to detect NO in aqueous solution and biological samples sensitively and selectively. METHODS Nitric oxide sensor was prepared by a 100μm diameter platinum electrode modified with a thin layer of chitosan, nickel sulfate and Nafion successively. The electrochemical reactions of NO on the electrodes were studied. The electrodes were used to determine NO released in rats' hippocampal slices by amperometry. RESULTS NO could be oxidized catalytically on the surface of the modified electrodes. The oxidation currents of NO on the electrodes showed a linear relationship to the concentration of NO in the ranger of 4.5×10 -4 ~6.7×10 -8 mol/L with the detection limit of 3.0×10 -8 mol/L. The electrodes were highly selective against ascorbate (200∶1), dopamine (200∶1) and nitrite (250∶1). CONCLUSIONS The electrodes were suitable for the determination of NO in vitro or vivo for their higher mechanic strength and high sensitivity to NO.
