摘 要:本研究旨在构建人H5N1流感病毒NA基因的重组腺病毒候选疫苗株,并考察其在BALB/c小鼠体内的细胞免疫效果。本研究选用流感病毒A/Anhui/1/2005(H5N1)的NA基因为研究对象,分别将野生型和按照人密码子优化的NA基因插入重组腺病毒载体,构建并鉴定了两种重组腺病毒rAdV-WtNA和rAdV-Mod.NA,纯化后病毒在第0周和第4周以肌肉注射方式免疫BALB/c小鼠两次,免疫剂量是109TCID50/只/次,第5周时使用Elispot方法检测并比较病毒的细胞免疫效果。结果显示,所制备和纯化的两种重组病毒均可有效表达NA目的蛋白;免疫后的小鼠均可检测出明显的针对NA抗原的特异性细胞免疫反应,而且rAdV-Mod.NA组分泌IFN-γ细胞的数量显著高于rAdV-WtNA组(P=0.016)。说明rAdV-Mod.NA是较好的人H5N1流感病毒候选疫苗株,值得进一步深入研究。The aim of this study is to develop the recombinant adenovirus vaccine (rAdV) candidates containing neuraminidase (NA) gene of H5N1 influenza virus and test in BALB/c mice the effect of cell-mediated immunity. In this study, two kind of NA gene (WtNA gene, the wild type; Mod. NA gene, the codonmodified type) derived from H5N1 influenza virus(A/Anhui/1/2005)were cloned and inserted respectively into plasmid of adenovirus vector, then the rAdV vaccines candidates (rAdV-WtNA and rAdV-Mod. NA) were developed and purified, followed by immunization intramuscularly (10^9 TCIDs0 per dose, double injection at 0 and 4tb week) in BALB/c mice, the effect of cell-mediated immunity were analysed at 5th week. Results indicated that: (i) NA protein expression was detected in two rAdV vaccines candidates by Western blotting; (ii) the rAdV-Mod. NA vaccine could elicit more robust NA specific cell-mediated immunity in mice than that of rAdV-WtNA vaccine (P=0. 016) by IFN-7 ELIspot assay. These findings suggested rAdV-Mod. NA vaccine was a potential vaccine candidate against HSN1 influenza and worthy of further investigation.
