摘  要:目的研究诊断超声照射大鼠早孕胚胎组织细胞Bcl-2表达的影响,探讨诊断超声诱导细胞凋亡的机制。方法采用频率为3.5MHz超声分别照射孕7d大鼠腹部子宫体表投影区,分为照射0min组、10min组、20min组、30min组,超声照射后24h取胚胎组织,SP免疫组织化学检测Bcl-2蛋白表达水平的变化。结果随着照射时间的延长,各组Bcl-2阳性表达率逐渐下降,与照射时间呈负相关。其中10min组与0min组相比差异无显著性(P>0.05),20min组、30min组与0min组相比差异有非常显著性(P<0.01)。结论诊断超声持续照射大鼠早孕胚胎超过20min使Bcl-2蛋白水平表达下调,提示诊断超声长时间照射能抑制抗凋亡因子Bcl-2的表达,从而诱导细胞凋亡。Objective To investigate the influence of Bcl-2 protein expression on rat embryo cells in the first tri mester pregnancy induced by diagnostic ultrasound exposure and its apoptosis mechanism. Methods Early pregnant SD rats were irradiated by sonography (frequency 3.5 MHz) and divided into four groups: group 0 min, 10 min, 20 min and 30 min. Changes of rat embryo cells Bcl-2 protein expression level were detected by SP-immunohistochemical technique. Results The rate of positive expression has been gradually declining with the extension of exposure time. There was no significant difference between group 10 min and 0 min (P〉0.05), but there were significant differences among group 30 min, 20 min and 0 min (P〈0.01) . Conclusions The Bcl-2 protein expression level goes down with the exposure time, which suggests that diagnostic ultrasound exposure can inhibit the anti-apoptotic factor Bcl-2 expression and thereby induce cell apoptosis.
