摘 要:为比较99mTc-AMLA的特异性,将两组心肌梗塞24h大鼠分别注射99mTc-AMLA(n=24)和99mTc-N-IgG(n=18);为研究不同心肌梗塞时间对摄取抗体的影响,将另一组不同心肌梗塞时间(2h到14d共8个亚组)的大鼠注射99mTc-AMLA,计算三组的ID%/g和梗塞心肌摄取/正常心肌摄取比(IM/NM)。用一只心肌梗塞的狗注射99mTc-AMLA后离体心肌显像。结果表明,梗塞心肌摄取99mTC-AMLA为特异性摄取。其摄取量远高于正常心肌的摄取。大鼠注射99mTc-AMLA8h后IM/NM最高为6.68;而注射非特异性IgG在相同时刻仅为1.42(P<0.01)。大鼠梗塞心肌摄取99mTc-AMLA与梗塞时间无明显区别(P>0.05)。离体狗心肌显像表明,心肌中放射性浓集区与TTC染色中空白区一致。说明99mTc-AMLA是一个对心肌梗塞特异的显像剂,它可用于亲心肌梗塞的显像。The feasibility and specificity of imaging of myocardial infarct with 99mTc-AMLA was reported. The first group of myocardial infarction(MI) rats were injected 99mTc-AMLA at 24h post MI. The second group were injected with 99mTc-N-IgG for control. For study of the influence of MI onset to uptake of 99mTc-AMLA, the third group with varied duration of MI from 4h to 14d was used.ID%/g and Infracted/Normal uptake ratio(IM/NM) were calculated. One in vitroheart of MI dog was imaged at 24 h after injection of 99mTc-AMLA. The results showed that the infarcted myocardium uptake of 99mTc-AMLA was higher than the normal myocardium. The IM/NM reached 6.68 at 8h in the first group, while non-specific IM/NM of 99mTc-N-IgG was only 1.42 (P<0.01). There were no significant differences between the ratios of uptake of 99mTc-AMLA in any infarcted duration from 4h to 14d(P>0.05). The in vitro myocardium imaging showed that the localization of 99mTc-AMLA in myocardium was the same as the blank area of triphenyltetrazolium chloride staining. It was concluded that 99mTc-AMLA is a myocardial imaging agent specific to MI. Thereby, it can be used as a useful infarct avid imaging reagent.
