摘 要:目的:建立LC-ESI-MS法测定血浆中盐酸雷尼替丁的血药浓度。方法:采用Zorbax SB-C18(2.1×100 mm,3.5μm)色谱柱,流动相为乙腈-0.1%甲酸溶液(30:70)。电喷雾离子源(ESI),选择正离子模式检测,采用选择离子监测(SIM)方式进行定量分析,检测离子为m/z 315(雷尼替丁),m/z 180(内标非那西丁)。结果:血浆中盐酸雷尼替丁浓度在5~1000 ng/ml范围内线性关系良好,最低检测限为5 ng/ml。盐酸雷尼替丁的提取平均回收率为83.92%~103.5%,日内及日间RSD都低于12%。结论:该法操作简便、快速、灵敏度高,可用于雷尼替丁的临床血药浓度监测。Objective:A LC-ESI-MS method was established for determination of ranitidine hydrochloride in human plasma.Methods:A Zorbax SB-C18(2.1×100 mm,3.5 μm) column was used,with mobile phase of acetonitrile-0.1% formic acid(30:70).Electrospray ionization(ESI)source was applied and operated in positive ion mode.Selected-ion monitoring(SIM)mode was used.The detected ions were m/z 315(Ranitidine),m/z 180(internal standard,Phenacetin).Results:The calibration curve of ranitidine hydrochloride was linear in the concentration range of 5~1000 ng/ml.The LOD was 5 ng/ml.The average extraction recovery was 83.92%~103.5%.The RSDs of inter-day and intra-day were less than 12%.Conclusion:The analytical method for ranitidine hydrochloride is accurate,sensitive and suitable for clinical blood concentration monitoring.
