摘  要:目的:观察银杏叶提取物对慢性马兜铃酸肾病(CAAN)大鼠肾组织纤维化的影响。方法:大鼠30只予关木通水煎剂灌胃12周,制备大鼠马兜铃酸肾病模型,12周后随机分成未治疗组和治疗组。治疗组(n=15)大鼠予银杏叶〔银杏天宝片,300mg/(kg.d)〕混悬液灌胃8周,未治疗组(n=15)予饮用水灌胃8周。另设健康对照组(n=10)。实验20周时处死所有大鼠,留取血、尿、肾组织标本,行血红蛋白、肾功能、尿蛋白、病理、免疫组化检查及逆转录聚合酶链反应(RT-PCR法)测定纤维化相关因子。结果:与未治疗组比较,治疗组血尿素氮(BUN)、肌酐(Scr)明显下降(P<0.05);肾间质纤维化程度减轻;免疫组化结果:治疗组肾组织转化生长因子β1(TGF-β1)、结缔组织生长因子(CTGF)、纤维连接蛋白(FN)光密度值较未治疗组明显下降(P<0.05);逆转录聚合酶链反应(RT-PCR法)测定肾组织TGF-β1、CTGF的mRNA表达,结果治疗组均明显减低(P<0.05)。结论:银杏叶提取物能够减轻大鼠CAAN肾组织纤维化。Objective : To observe the effect of Folium Ginkgo on fibrosis aristolochic acid nephropathy. Methods : The rat model of aristolochic acid nephropathy was induced by producing Caulis aristolochiae manshuriensis for 12 weeks. 30 model rats were randomly divided into 2 groups : the therapy group and non - therapy group. The therapy group ( 15 ea- ses) was treated with Folium Ginkgo [ 300mg/( kg· d) ] orally for 8weeks. The non - therapy group ( 15 cases) was given water. Anothe 10 rats of the change of BUN, serum creatinine were observed respectively after 8 weeks. The control group were given water togethe. Renal histology was examined and the mRNA of TGF - β1 and CTGF were reversely tran- scribed and quantified to real - time PCR. Results : Compared with the non - therapy groups, the levels of BUN and Scr were significantly lower in therapy group (P 〈 O. 05, respective). Renal interstitial fibrosis in therapy group was less seri- ous than that of the non - therapy groups. The optical density of TGF - β1, CTGF and Fn with therapy group was lower than that of the non - therapy groups. On the contrary, the expression of TGF -β1 and CTGF mRNA was down - regulated in therapy group than that of in non - therapy group ( P 〈 0.05). Conclusion: Folium Ginkgo can alleviate the fibrosis and protect the renal function of aristolochic acid nephropathy.
