摘 要:目的:观察体外高糖培养条件下,消可宁颗粒对血管内皮细胞内皮型一氧化氮合酶(endothelial nitric oxide synthase,eNOS)、诱导型一氧化氮合酶(induce nitric oxide synthase,iNOS)、一氧化氮(nitric oxide,NO)及过氧亚硝酸阴离子(peroxynitrite,ONOO-)的影响。方法:体外培养人脐静脉内皮细胞株ECV-304,分常规培养组、高糖对照组和不同浓度消可宁组,检测NOS和NO的吸光度,计算出活力或含量。用免疫组织化学方法检测ONOO-生成的标记物一硝基酪氨酸(nitrotyrosine,NT)的表达。结果与高糖对照组比较,消可宁组eNOS活力明显增加,iNOS活力降低,NO含量下降,NT表达减少。并具有剂量与时间依赖性。结论:消可宁可以抑制iNOS活性,并降低NO含量,减少血管内皮细胞内ONOO-形成而发挥抗氧化作用。Objective:To explore the effects of Xiaokening in high glucose on nitric oxide synthase (NOS), nitric oxide (NO) and peroxynitrite (ONOO-) produced by vein endothelial cells. Methods:Human umbilical vein endothelial cells strain ECV -304 were divided into seven groups:normal groups:high -glucose group (30mM) and the Xiaokening groups (20,40,60,80,100g/L). The eNOS, iNOS, NO and nitrotyrosine (NT) were detected at the end of the experiment. Results : Compared with high - glucose group, eNOS activity increased in Xiaokening groups, ( P 〈 0.05 ). The change in 100g/L was greater than that in 20g/L (P 〈 0.05). While iNOS, NO and NT decreased. Conclusion: Xiaokening can take more effect on inducing eNOS and inhibiting iNOS,NO and ONOO - release from vein endothelial cells than that of high glucose.
