摘 要:目的:探讨肾脏水通道蛋白2(AQP2)的表达与慢性肾衰大鼠水液代谢异常及右归丸防治作用的机制。方法:采用腺嘌呤灌胃法建立慢性肾衰大鼠模型,将实验动物随机分为正常对照组,模型组,右归丸组。采用免疫组化、western blot方法检测各组大鼠肾脏中AQP2的表达,并测定各组大鼠24h尿量、尿比重、血Scr及BUN水平。结果:与正常对照组相比,模型组大鼠尿量明显增加,尿比重下降,血Scr及BUN水平显著升高,肾脏AQP2的表达显著降低(P<0.05);与模型组比较,右归丸组大鼠尿量减少,尿比重增加,血Scr及BUN水平显著下降,肾脏AQP2的表达显著增加(P<0.05)。结论:模型组大鼠肾组织中AQP2表达下调可能参与慢性肾衰水液代谢紊乱的形成,右归丸对AQP2的表达有上调作用,其可能通过上调AQP2表达,纠正慢性肾衰水代谢紊乱,改善肾功能,延缓慢性肾衰的进展。Objective:To observe the relationship between expression of AQP2 and abnormal fluid metabolism in chronic renal failure and to explore the mechanism of Yougui Pill in the treatment of chronic renal failure. Methods : CRF model was induced by Adelne ig. The rats were randomized into normal group, CRF model group, yougui Pill treatment group. Change of AQP2 expression in the kidney of rats in each group were analyzed by immunohistochemistry, and western blot. Urine volume, Urine osmolality, serum ereatinine and urea nitrogen levels were measured. Results:Compared with normal group, Urine volume increased and Urine osmolality decreased in CRF model group, Serum creatinine and urea nitrogen levels were higher, AQP2 expression in the renal tissue of CRF model group decreased significantly ( P 〈 0.05 ). While AQP2 expression in yougui Pill treatment group was higher than in CRF model group( P 〈 0.05 ). Urine volume decreased and Urine osmolality increased as compared with that in the model group. Serum creatinine and urea nitrogen levels were lower. Conclusion:The expression of AQP2 decreases in the renal tissue of CRF rat model,and the decrease might participate in the formation of abmormal fluid metabolism in CRF. Yougui Pill may have beneficial effects on CRF by regulating AQP2 expression in the kidney of rats.
