摘  要:目的观察1型糖尿病大鼠肾脏衔接蛋白p66Shc的表达及活性氧的水平。方法以链脲佐菌素制备糖尿病大鼠模型,分为糖尿病组和正常对照组。采用双缩脲法检测24h尿蛋白;免疫组化、Westem blot方法检测肾脏p66Shc表达;二氢乙啶染色检测肾组织活性氧。结果与对照组相比,糖尿病大鼠24h尿蛋白明显增高(P〈0.01),肾脏特别是肾小管p66Shc的表达显著增高(P〈0.05),活性氧含量明显增高。结论糖尿病大鼠肾脏p66Shc表达增强,活性氧含量增加,p66Shc可能在糖尿病肾病肾组织氧化损伤的发生发展过程中扮演重要角色。Objective To observe the expression of adaptor protein p66Shc and the level of reactive oxygen species in the kidneys in STZ-induced diabetic rats. Methods Diabetes was induced by the intrapefitoneal injection of STZ. 24h urine protein was quantitated by biuret method. The expression of p66Shc was examined by immunohistoehemically staining and Western blot, respectively. The level of reactive oxygen species was detected by DHE staining. Results Compared with control, urine protein in diabetic rats was increased ( P 〈 0. 01 ). The expression of p66Shc was significantly increased, especially in renal tubule in diabetic rats, compared to control group ( P 〈 0.05 ). Severe oxidative stress was also observed in kidneys of diabetic rats. Conclusion The adaptor protein p66Shc expression was significantly upregulated and it was related with increase of ROS level in rat kidneys with diabetes, which suggested that p66Shc might be involved in the process of oxidative injury in diabetic nephropathy.
