摘  要:目的探讨核因子-κB(NF-κB)在成人原发性肾病综合征(PNS)发病机制中的意义,研究PNS患者外周血单个核细胞(PBMC)中NF-κB活性对白细胞介素-8(Interleukin-8,IL-8)水平的调控作用。方法以28例确诊PNS的成人患者(PNS组)和25例健康成人(对照组)为研究对象,抽血、提取PBMC并培养,用酶联免疫吸附法分别检测2组的NF—κB活性吸光度(OD)值及酶联免疫吸附双抗体夹心法检测IL-8的浓度。再分别用NF—κB的特异性激动剂IL-1β和特异性抑制剂TPCK(N—tosy1 Phenylalanine Chlormethy1 Ketone)进行干预后,分别检测两组的NF—κB活性吸光度和IL-8的浓度。结果与正常对照组相比,PNS组NF-κB活性及IL-8浓度均显著增高,差异有统计学意义(P〈0.05),且随着NF—κB活性的增强和抑制,IL-8的浓度也相应升高和降低,两者有直线相关关系(P〈0.05),而正常对照组无此相关关系。结论NF—κB在成人原发性肾病综合征发病中有重要意义,可能是通过调节IL-8的浓度而发挥其致病作用。Objective To investigate the relationship of pathogenesis in adults'primary nephritic syndrome (PNS) with PBMC nuclear factor kappa (NF-κB) and study the adjustments of IL-8 caused by active PBMC NF-κB in the patients with PNS. Methods 28 patients with PNS and 25 normal adults were enrolled in this study. PBMC were isolated and separated from fresh blood in asepsis condition, and cultured by RPMI 1640. It was divided into 3 groups. Control group was normally cultured, group IL-1β was cultured with IL-1β, and group TPCK was reacted with inhibitor of TPCK. The activity of NF-κB and concentration of IL-8 in supernatant were measured by ELISA. Results The activity of PBMC NF-κB and concentration of IL-8 of PNS group were significantly higher than that in the control group( P 〈 0. 05 ). The activity of PBMC NF-κB of PNS group showed linear correlation with the level of IL-8 ( P 〈 0. 05 ). Conclusion The levels of IL-8 in patients with PNS may be pathological intervened with NF-κB. NF-κB and IL-8 may play an important role in the pathogenesis of PNS.
