摘 要:目的:观察葛根素能否在体外减少葡萄糖腹膜透析液中晚期糖基化终末产物(AGEs)的产生。方法:将20 g牛血清白蛋白充分溶解于1 000 m l 2.5%腹膜透析液中,根据葛根素终浓度分5组观察(葛根素终浓度A组为0 g.L-1,B组为0.01 g.L-1,C组为0.05 g.L-1,D组为0.1 g.L-1,E组为0.2 g.L-1)。过滤除菌后37℃保存,分别于第1天、7天、1个月、2个月以及3个月时取样本1 m l进行AGEs测定。结果:腹膜透析液中添加牛血清白蛋白后,随着时间的延长,透析液中AGEs的浓度逐渐升高。加入不同浓度葛根素的试验组,其AGEs的浓度均较同期未加入葛根素的试验组明显降低,并且随着葛根素浓度增加,AGEs的浓度渐降。结论:葛根素对体外腹膜透析液中AGEs形成有明显的抑制作用,且它的这种抑制作用呈现剂量依赖性。Objective To investigate the effect of Puerarin on the formation of advanced glycosylation end products (AGEs) derived from commercial peritoneal dialysis solution (PDS) in vitro. Methods 20 g fetal bovine serum albumin and various dosage of Puerarin were thoroughly dissolved in 1 000 ml PDS containing 2.5 % glucose. Five groups which final concentrations of Puerarin were 0,0.01,0.05,0.1 and 0.2 g · L^-1 were included in this study. After being filtersterilized, they were incubated in 37 ℃. And then, 1 ml solution got from each group were frozen for AGEs deteetion at the day 1,7 and month 1,2,3 respectively. Results As time went on, the amount of AGEs generated in the PDS were escalated in all five groups. While, at the same time point, evidentless AGEs could be detected in the groups with higher eoneentration of Puerarin than that in the groups with lower concentration of Puerarin. Conclusion In vitro, Puerarin may induce a dose-dependent inhibition of the formation of AGEs in PDS.
