摘  要:目的:探讨人参皂甙Rg1对大鼠室管膜前下区神经干细胞(SVZa NSCs)的保护作用及其与STAT3关系。方法:采用体外细胞培养法对胚胎大鼠SVZa NSCs分离、培养,建立SVZa NSCs谷氨酸中毒模型,相差显微镜下观察细胞形态变化,用台盼蓝排斥试验和MTT试验检测存活率,用TUNEL试验检测凋亡率;用免疫细胞化学法检测STAT3阳性细胞百分比表达。结果:人参皂甙Rg1能够明显提高谷氨酸损伤SVZa NSCs的存活率;可以提高损伤细胞STAT3阳性细胞百分比的表达。结论:人参皂甙Rg1对谷氨酸兴奋性中毒的SVZa NSCs有保护作用,其作用机制可能与增加STAT3表达有关。Objective:To research on Ginsengnoside Rgl on Enhancing Protection of SVZa-NSCs of Rats and Expression of STAT3. Methods : Developing the Glu poison model of rat SVZa-NSCs by means of embryo in vitro to observe changes of the cells under microscopes, using trypanbluc tests and MTT tests to detect the living probability, detecting dying probability by means of TUNEL experiment, detecting STAT3 expressions and the percentage of positive cells using immunocytochemistry method, discussing the protecting functions of NSCs by Rgl. Results:Rgl have obvious functions in increasing surviving of Glu poisonous SVZa-NSCs and increasing of STAT3 expressions and the percentage of positive cells. Conclusions : Rgl have obvious anti-Glu poisonous functions and its mechanism may be linked with the increasing of STAT3 expressions.
