摘 要:目的:探讨玉竹乙醇提取物对心梗后心衰大鼠血流动力学的影响并进行量效关系的研究。方法:70只清洁级健康成年SD大鼠随机分为5组,即假手术组,模型组,玉竹大、中、小剂量组,造模成功后尾iv不同剂量玉竹乙醇提取物,测定用药后不同时点(0,5,10,20,30 min)大鼠心率(HR),左室收缩压最大压(LVSP),左室舒张末压(LVDP),左心室压力最大上升速率(+dp/dtmax),左心室压力最大下降速率(-dp/dtmax),并分别进行组内及组间相同时段的比较。结果:玉竹大、中剂量组用药5min后开始HR减慢、LVSP明显下降,+dp/dtmax、-dp/dtmax减慢,LVDP明显上升,20-30 min以后基本恢复正常,小剂量组只在10 min时出现明显改变,并且HR未见明显变化。结论:玉竹具有负性肌力、负性频率作用,并随剂量增大呈现出一定的量效关系;小剂量时负性肌力为主,随着剂量增大,负性肌力作用增强,同时出现负性频率作用。Objective: Objective:To observe the effects and dose-effect relationship of "ethanol extract from Polygonatum Odoratum" on Hemodynamics in Rats of Heart Failure Following Myocardial Infarction. Methods: Seventy healthy Sprague-Dawley rats were divived randomly into sham group, model group, large dosage of "ethanol extract from Polygonatum Odoratum" group, moderate dosage of "ethanol extract from Polygonatum Odoratum" group, small dosage of "ethanol extract from Polygonatum Odoratum" group. After successful model duplication, rats in different dosage of "ethanol extract from Polygonatum Odoratum" groups were treated with intravenous injection of 6 g/kg, 3 g/kg,0.3 g/kg distinctly, while rats in sham group or model group received 6 ml/kg saline. The heart rate(HR), left ventricular systolic pressure (LVSP), left vcntricular diastolic pressure ( LVDP), maximum rise speed of left ventricular pressure ( + dp/ dtmax), maximum drop speed of left ventricular pressure (-dp/dtmax)were measured at different time points (before injection and 5,10,20,30 minutes after injection)and comparatively analyed. Result:The HR decreased at 5 minutes in large dosage group and model group, the LVSP was lower, the + dp/dtmax and -dp/dtmax were slower, the LVDP increased at the same time, all these parameter went back to normal at 20 and 30 minutes points, the small dosage group was found changes in these parameters at 10 minutes point except HR. Conclusion: "ethanol extract from Polygonatum Odoratum" has negative inotropic and negative chronotropic action in CHF rats. These two actions have dose-effect relationship.
