Correlation Between ^(31)P-MRS with Desmin Expression in Skeletal Muscle of SD Mice after Exhaustive Swimming

摘  要:目的研究不同负荷游泳运动后SD大鼠骨骼肌31P-MRS的变化,以及与Desmin表达的相关性。方法将45只健康雄性SD大鼠随机分成3组:正常组(C组)、半力竭游泳组(HE组)、力竭游泳组(E组),麻醉后进行右下肢骨骼肌31P-MRS采集,测量Pi、PCr、ATP峰下面积并计算Pi/PCr、Pi/ATP、PCr/ATP。扫描后立即处死取材,常规组织学检查,并用免疫组化法进行Desmin染色,测算平均光密度值(OD值)。结果C组Pi、PCr、ATP峰下面积分别为0.16±0.05、1.33±0.62、0.41±0.06,Pi/PCr、Pi/ATP、PCr/ATP分别为0.14±0.08、0.39±0.13、3.28±1.62。光镜下,肌细胞形态规整,排列整齐。OD值为0.55±0.04;HE组Pi、PCr、ATP峰下面积分别为0.33±0.16、0.90±0.20、0.79±0.35,Pi/PCr、Pi/ATP、PCr/ATP分别为0.40±0.17、0.54±0.40、1.48±1.09。光镜下,HE组肌细胞水肿,肌丝分离,排列稀疏。OD值为0.32±0.01;E组Pi、PCr、ATP峰下面积分别为0.49±0.21、0.52±0.09、0.19±0.07,Pi/PCr、Pi/ATP、PCr/ATP分别为0.99±0.45、3.08±1.88、3.15±1.33。光镜下,E组部分肌丝断裂,皱缩。OD值为0.21±0.02;3组大鼠Pi、PCr、ATP及Pi/PCr存在组间差异性,Pi/ATP正常组与半力竭组无差异,PCr/ATP正常组与力竭组无差异。Pi、PCr、ATP、Pi/PCr、Pi/ATP与OD值相关系数依次为0.621、0.775、0.127、-0.75和0.583,均具有统计学意义(P<0.01)。PCr/ATP与OD值相关系数无统计学意义(P>0.05)。结论31P-MRS能够有效评价SD大鼠骨骼肌力竭性损伤,PCr峰下面积及Pi/PCr比值可作为检测骨骼肌力竭损伤的观察指标,且与病理分析具有相关性,因此能够定量定性分析骨骼肌损伤程度。Objective To evaluate the correlation of metabohc changes in 31p-MRS and Desmin expression in skeletal muscle of SD mice after exhaustive swinuning. Methods 45 SD male mice were randomly divided into 3 groups:control group (Group C ), half exhaustive swimming group(Group HE) and exhaustive swimming group(Group E). 31p-MRS was performed in right lower limb under anesthesia. MRS observation indexes included the peak areas of Pi, PCr, ATP and the ratios of Pi/PCr, Pi/ATP, PCr/ATP. The morphological changes were confirmed by histopathology, and imnmnohistocbernical methods were taken for the assessment of Desrnin expression with optical density( OD ) value. Results C group Peak areas of Pi,PCr,ATP are 0.16±0.05,1.33±0.62,0.41±0.06,ratios of Pi/PCr,Pi/ATP,PCr/ATP are 0.14± 0.08,0.39±0.13,3.28±1.62. Both morphology and arrangement of skeletal muscle cells were regular under microscope, OD value was0.55 ± 0.04;HE group Peak areas of Pi,PCr,ATP are 0.33±0.16,0.90±0.20,0.79±0.35,ratios of Pi/PCr,Pi/ATP,PCr/ATP are 0.40±0.17,0.54± 0.40,1.48±1.09. The disorder arrangement of skeletal muscle cells with edema and filaments separation were found under microscope,OD value was 0.32±0.01 ;E group Peak areas of Pi,PCr,ATP are 0.49±0.21,0.52±0.09,0.19±0.07,ratios of Pi/PCr,Pi/ATP,PCr/ATP are 0.99±0.45,3.08±1.88,3.15± 1.33, part of muscle filaments breakdown and atrophy were found under microscope, OD value was 0.21±0.02; The peak areas of Pi,PCr,ATP and the ratio of Pi/PCr were at a high level of significance among three groups (P 〈 0.01 ). The correlation eoefficients of Pi, PCr, ATP, Pi/PCr, Pi/ATP were 0.621,0.775,0.127,-0.75 and 0.583 respectively. There was no significance between PCr/ ATP and OD value (P 〉 0.05 ). Conclusion 31P-MRS eotdd quantitatively refleet the SD mice sport-induced injury after exhaustive exercise,which corresponding to morphological changes and histological structure damages.The peak area of PCr and the ratio of Pi/PCr which could evaluate the degree of
