摘 要:目的建立大鼠动脉硬化模型并研究内皮-氧化氮合酶基因(eNOS)在模型鼠股-腘动脉中的表达。方法在大鼠股-腘动脉内注入蒸馏水,通过低渗造成动脉内膜非机械性、浅表性损伤,联合高脂、高胆固醇等饲料饲养的方法,建立大鼠动脉硬化闭塞模型。术后15,30,90d,应用免疫组化技术观察动脉闭塞的程度,应用SYBR染料法荧光定量PCR技术分别检测模型鼠下肢股-腘动脉中eNOS基因的表达。结果术后大鼠血管内膜增厚、部分管腔闭塞。高脂+损伤后15d、30d和90d的大鼠股-腘动脉中eNOS基因的相对表达量分别为0.7219±0.0417、0.6444±0.0129和0.4601±0.0276(P<0.05)。结论成功建立大鼠动脉硬化闭塞模型。eNOS基因在模型鼠的下肢股-腘动脉中表达下调。Objective To construct rat arteriosclerosis obliterans (ASO) models and explore the expression of eNos in their femoral-pop- titeal artery. Methods After the endothelial injury of the rat femoral-popliteal artery was performed by distilled water injection surgery, the rats were fed with high-fat,high-cholesterol diet,and thus the rat ASO models were established. Using immunohistochemistry and SYBR- Green I PCR technique ,we observed the arterial occlusion level and the expression of eNOS in femoral-poptiteal artery of the rat ASO models and the normal conm31s on 15,30,90 days after surgery. Results Typical pathological changes of ASO, such as connective tissue prolif- eration and arrangement disorder, lumens stenosis ,thrombosis, foam cells in plaques, visible necrosis and calcification foci formation were observed. The relative expression levels of eNOS in the femoral-poptiteal artery of model rats were 0.7219±0.0417 ( 15 days after surgery), 0.6444±0.0129 (30 days after surgery),0.4601±0.0276 (90 days after surgery) respectively. The eNOS expression levels on the 3 time points were different significantly (P 〈 0.05). Conclusion The rat ASO model can be established by femoral-popliteal arterial endothelial injury plus high-fat, high-cholesterol diet. The expression levels of eNOS in the femoral-poptiteal artery decreased in the ASO model rats.
