摘 要:目的探讨超声观察兔腹主动脉粥样硬化斑块的形成过程,以及炎症对其的影响。方法将18只日本大耳兔随机分成对照组、高脂组、炎症组;对照组喂饲普通饲料,高脂组及炎症组均喂饲高脂饲料建立动脉粥样硬化模型,炎症组同时腹腔注射酵母多糖A,喂养12周。于实验开始第0周及第12周末分别对3组家兔进行超声检查,观察腹主动脉内中膜厚度(IMT)改变及斑块形成情况。12周后处死各组家兔取其腹主动脉行病理学检查。采用RT-PCR法检测腹主动脉壁中CRP mRNA的表达情况。结果与高脂组比较,炎症组腹主动脉CRP mRNA表达显著增高,IMT显著增厚。超声检查结果与病理学检查结果吻合一致。结论炎症反应能加速动脉粥样硬化形成的进程。超声检查为评价动脉粥样硬化提供一种简便、可靠、经济、实用的检测方法。Objective To observe the formation process of atherosclerosis plaque and the action of inflammatory reaction with the ultrasound technology in rabbits. Methods Eighteen Japanese white rabbits were divided into three groups randomly,i, e. control group,high fat diet group and inflammation group. All the animals were fed with high-cholesterol diet except that in control group. Rabbits in inflammation group were administrated intraperitoneally with zymosan A 10mg/kg, once every two days. The initial-medial thickness (IMT) of abdominal aortic posterior wall and the plaque were measured with ultrasound technique before and after 12 weeks feeding. And the expression level of C-reactive protein mRNA was detected with RT-PCR method. Animals were killed after 12 weeks and abdominal aortas were prepared for pathology exam. Results The IMT in inflammation group increased significantly compared with high fat diet group (P〈0.01). The results of ultrasound were consistent with pathological examination. Simultaneously, the expression level of C-reactive protein mRNA increased significantly. Conclution The inflmmation could accelerate the process of AS. Ultrasound technique is a simple and reliable method to evaluate its action on atherosclerosis.
