三七皂苷对KK-Ay鼠的降糖减肥及肾病变保护作用

摘　　要：目的:采用遗传性2型糖尿病(DM)鼠考察三七皂苷(PNS)的降糖、减肥及肾病变保护作用。方法:C57BL/6J健康鼠及KK-Ay鼠,分为正常对照组、模型组及PNS低(50 mg/kg)、高(200 mg/kg)剂量组共4组,腹腔注射给药30 d,观察PNS治疗对小鼠空腹血糖水平(FPG)、糖耐量、体质量、摄食量、血清胰岛素、胰岛素抵抗指数(IRI)、甘油三酯(TG)、胆固醇(CH)及肾小球损伤的影响。结果:PNS高剂量治疗第12、22、30天KK-Ay鼠的FPG及体质量增长率显著降低,12 d后的糖耐量有显著改善,30 d后的IRI、TG含量显著降低,肾小球损伤得到显著抑制,各作用均呈量效关系。结论:PNS对2型糖尿病KK-Ay鼠具有降糖、减肥和抑制肾病变作用。Objective：To observe the effects of Panax notoginseng saponins（PNS） on anti-hyperglycemic, anti-obese and prevention from kidney pathological changes in a type 2 diabetic KK-Ay gene mice model. Methods：All animals were divided into 4 groups： Normal control group（ C57BL/6J mice）were treated by NS 10 mL/kg, KK-Ay mice were subdivided into 3 groups as DM administrated NS 10 mL/kg, PNS 50 mg/kg, PNS 200 mg/kg respectively. All the animals received daily intraperitoneal injections for 30 days consecutively. All of these following items were determined as the effect of PNS：fasting plasma glucose levels （FPG） ,intraperitoneal glucose tolerance, body weight, food intake, insulin resistance index （ IRI）, serum insulin, triglyceride （ TG）, cholesterol （ CH ） levels and contribution of glomcrulus injury. Results：On the 12 th,22 th and 30 th day, PNS-treated group had significantly lower FPG and low body weight incremental percentage. After a 12-day treatment, glucose tolerance was significantly improved. On the 30th day the serum IRI and TG levels of PNS-treated group decreased significantly, and the development of the mice glomerular lesions was prevented sig- nificantly. All of these showed a dose-effect relationship. Conclusion ： PNS has the effects of anti-diabetes, anti-obese and prevention from kidney pathological changes in type 2 diabetic KK-Ay mice.
