摘 要:目的比较益气活血方(脑络欣通)和补肾生髓方对脑缺血再灌注大鼠缺血侧侧脑室下区神经上皮干细胞蛋白(Nestin)、缺血半暗带神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)表达的影响。方法采用线栓法制作局灶性脑缺血再灌注动物模型,动物随机分为假手术组、模型组、益气活血方组及补肾生髓方组。脑缺血2h,分别再灌注1周、2周、3周,采用免疫荧光法检测Nestin、NSE和GFAP表达的阳性细胞数或平均吸收光密度值(A值)。结果缺血侧侧脑室下区,再灌注1周、2周益气活血方组,再灌注2周、3周补肾生髓方组,Nestin表达的阳性细胞数较模型组显著增多(P<0.05或P<0.01);再灌注1周益气活血方组较补肾生髓方组显著增多(P<0.01),再灌注2周、3周补肾生髓方组较益气活血方组显著增多(P<0.05)。缺血半暗带区,再灌注1周、2周益气活血方组,再灌注2周补肾生髓方组,NSE表达的阳性细胞数较模型组显著增多(P<0.01);再灌注1周时益气活血方组较补肾生髓方组显著增多(P<0.01),再灌注2周时补肾生髓方组较益气活血方组显著增加(P<0.01)。缺血半暗带区,再灌注1周、2周益气活血方组与补肾生髓方组GFAP表达平均吸光度较模型组明显增加(P<0.05或P<0.01);再灌注各时间段两复方组比较无显著差异(P>0.05)。结论益气活血方与补肾生髓方能通过增加缺血侧侧脑室下区Nestin、缺血半暗带区NSE数量、增强GFAP的表达,促进局灶性脑缺血后神经干细胞的增殖分化,两种复方在增加Nestin、NSE数量方面存在一定差异。Objective To compare the influences of Yiqihuoxue Formula (using Naoluoxintong, YF) and Bushenshengsui Formula (BF) on the expressions of Nestin in the subventricular zone (SVZ) of ischemie lateral ventricle, neuron specific enolase (NSE) and glial fibrillary acidic protein (GFAP) in the isehemic half-dark zone in rats with focal cerebral ischemia-reperfusion (I/R). Method The rat model of focal cerebral I/R was established by using thread approach, and then the rats were randomly divided into the sham-operation group, model group, YF group and BF group. After cerebral ischemia for 2 hours the reperfusion was given to the rats for 1 week, 2 weeks or 3 weeks respectively. The positive cell quantity and the value of average absorbance (A value) in the expressions of Nestin, NSE and GFAP were detected by using immunofluorescence staining method. Result In SVZ the positive cell quantity in Nestin expression was higher significantly in YF group after I/R for 1 week and 2 weeks, and in BF group after I/R for 2 and 3 weeks than that in the model group (P 〈0.05 or P 〈0.01 ), that in YF group after I/R for 1 week was higher significantly than that in BF group (P 〈0. 01 ), and that in BF group after I/ R for 2 and 3 weeks was higher significantly than that in YF group ( P 〈 0.05 ). In the ischemic half-dark zone the positive cell quantity in NES expression was higher significantly in YF group after I/R for 1 week and in BF group after I/R for 2 weeks than that in the model group ( P 〈0.01 ) that in YF group after I/ R for 1 week was higher significantly than that in BF group (P 〈0.01 ), and that in BF group after I/R for 2 weeks than that in YF group (P 〈0.01 ). In the ischemic half-dark zone the average absorbance of GFAP expression was higher significantly in YF group after I/R for 1 week and 2 weeks and in BF group than that in the model group ( P 〈 0.05 or P 〈 0.01 ) ).
