摘 要:目的:研究小檗碱对高迁移率族蛋白B1(HMGB1)胞外释放的影响及其机制。方法:观察小檗碱对内毒素脂多糖激活后培养巨噬细胞株RAW264.7HMGB1基因表达和释放的作用,及对内毒素血症小鼠血清HMGB1水平和存活率的影响。结果:小檗碱(100μmol/L)明显抑制内毒素脂多糖激活后巨噬细胞HMGB1的mRNA表达和胞外释放(P<0.05)。内毒素血症小鼠腹腔注射小檗碱(5mg/kg)后,血清HMGB1水平降低,96h后小鼠存活率由28%提高至56%。结论:小檗碱具有抑制细胞HMGB1释放和保护内毒素血症小鼠作用。Objective: To study the effect of berberine on extracellular release of high mobility group box 1 (HMGB1) and its mechanism. Methods: The effect of berberine on release of HMGB1 and expression of HMGB1 mRNA were investigated in cultured macrophage-like RAW 264.7 cells after stimulation with lipopolysaccharide. The effect of berberine on serum HMGB 1 level and survival rate of endotoxemia mice was also studyed. Results: Berberine(100pmol/L) markedly suppressed the release of HMGB1 and expression of HMGB 1 mRNA from lipopolysaccharide-stimulated macrophages(P 〈 0.05). Serum HMGB1 level decreased, and the survival rate elevated from 28% to 56% at 96 hours, after endotoxemia mice were injected with berberine(5mg/ kg) intraperitoneally. Conclusion: Berberine was an effective inhibitor of HMGB 1 release, and confered protection against endotoxin lethality.
