摘 要:本实验利用高效毛细管电泳建立了同时检测雏鸡、哺乳猪等饲料中5种氟喹诺酮类药物的方法。研究了缓冲液的类型、离子浓度、pH、分离电压和分离温度等条件对分离的影响,通过正交实验设计得到了最佳的电泳条件,在274nm处,分离电压为18kV,分离温度为22℃,在80mmol/L柠檬酸-柠檬酸钠pH6.5的运行缓冲液下,5种药物可在10min内完全分离。5种药物的浓度与峰面积的相关系数r=0.9999~0.9994,方法精密度(RSD)为1.36%~1.69%(迁移时间)和2.09%~5.89%(峰面积)。同时建立了饲料的前处理方法,方法平均回收率为84.42%~107.5%。该方法操作简便、准确、快速。High-performance capillary electricity (HPCE)was used for the determination of five fluoroquinolones in feeds.The effects of some important factors such as pH,composition and concentration of running buffer, separation voltage and temperature were investigated.The optimum condition was acquired by the orthogonal test design.In 274 nm wavelength, the five components could be well separated within 10 min at the separation voltage of 22 kV and temperature 22 ℃ in 80 mmol/L Citric acid-sodium citrate at pH 6.5.Their corresponding coefficient reached from 0.9999 to 0.9994. The precision of migration time was 1.36 % - 1.69 % and peak area was 2.09 % - 5.89 %.Pretreatment method of feeds was established.The average recoveries were from 84.42 % to 107.5 %.The method is rapid,simple and accurate.
