摘  要:目的研究体外培养的雪旺氏细胞VEGF分泌量与培养时间的关系,为今后雪旺氏细胞的应用提供实验依据。方法切取2周龄SD大鼠的坐骨神经,分离、纯化、培养雪旺细胞,第三代细胞分别于12小时,1天,3天,5天,7天,9天6个时间位点收集细胞以及细胞培养液,观察细胞形态变化,S-100鉴定雪旺细胞,同时,四氮唑溴盐(MTT)比色法测定雪旺细胞的生长增值情况;ELISA法测定细胞培养液中的VEGF浓度含量。结果雪旺细胞呈长梭形,S-100蛋白表达阳性率为90%,2~5天细胞生长迅速,7天以后生长较为缓慢,分泌的VEGF量在第5天达到最高(每个时间点均P<0.05),其后第7天,第9天缓慢降低(P>0.05)。结论第3代雪旺细胞在培养到第5-7天时达到较高的分泌浓度,这个时期的细胞在血管组织工程中有较好的应用价值。Objective To study secreting of vascular endothelial growth factor (VEGF) in cultured rat Schwann' s cells, and to provide evidence for the cell' s application in vascular tissue engineering. Method Schwann ceils were harvested from the 2-week-old SD rat sciatic nerves, separated, purified, cultured and passaged twice. The supematants of the third generation of Schwann' s cells, at 12h,1d, 3d, 5d, 7d, 9d were collected respectively for ELISA assay. The remained cells were checked morphologically, and identified with S-100 staining separately. Results The spindle-shaped cells overlaid in the flask, and more than 90% of the cells were positive for the s-100 staining. The VEGF level increased at 12h, ld, 3d, 5d, and then decreased at 7d, 9d respectively. Conclusions The cultured 3-5 days of the 3rd generation Schwann' s cells, with highest VEGF levels, were applicable in vascular tissue engineering.
