摘  要:目的:探讨川芎嗪是否可通过调节血管内皮生长因子(VEGF)表达而影响大鼠实验性关节炎的发展。方法:建立大鼠佐剂性关节炎(AIA)模型,灌胃给药川芎嗪,检测大鼠足垫厚度的变化,免疫组化SP法染色CD34检测滑膜微血管密度(MVD),放射免疫法分析大鼠血清中肿瘤坏死因子(TNF-α)的含量,Western blot分析滑膜组织VEGF的表达。结果:与模型组相比较,100 mg.kg-1川芎嗪组大鼠足垫厚度显著减小,血清中TNF-α的含量和滑膜组织VEGF的表达均明显降低。结论:川芎嗪抑制大鼠佐剂性关节炎的发展与抑制病变关节滑膜VEGF的表达密切相关。Objective: To explore the effect of tetramethylpyrazine (TMP) on the expression of vascular endothelial growth factor (VEGF) in the synovium of adjuvant-induced arthritis (AIA) in rats. Method: AIA rats were treated with different doses of TMP. The effects of treatment were monitored by footpad thickness, contents of tumor necrosis factor-α (TNF-α) in serum, microvessel density (MVD) and the expression of VEGF protein in the synovium. Result: Compared with arthritis model, 100 mg · kg^-1 TMP could remarkably reduce the footpad thickness, contents of TNF-α in serum and the expression of VEGF in the synovium. Conclusion: TMP can attenuate the degree of chronic inflammation in AIA rats, and its mechanism might be associated with inhibiting the expression of VEGF.
