丹参注射液给药时间对预防顺铂耳毒性作用的影响

摘　　要：目的探索丹参注射液给药时间对预防顺铂耳毒性作用的影响。方法健康杂色豚鼠40只。随机均分为4组。1．对照组：腹腔注射顺铂1mg／kg／d。2．实验Ⅰ组：腹腔注射顺铂1mg／kg／d，6h后腹腔注射丹参注射液8mL／kg／d。3．实验Ⅱ组：腹腔同时注射顺铂和丹参注射液，剂量同上。4．空白组：腹腔注射生理盐水0．8mL／kg／d。各组连续6d。各组动物在实验前及结束后各做一次BAEP检测。以ABRⅢ波反应阈为检测听阈。结果对照组与实验组听阈在实验前后同组内相比均有提高，分别为：42．6±4．32dB、33±3dB、36．6±2．16dB，空白组为31．5±2．25dB。但实验Ⅰ组和实验Ⅱ组听阈分别比对照组低，差别显著。实验前后同组内相比听阈增加率：实验Ⅰ组〈实验Ⅱ组〈对照组，分别为：（6±8．4）％、（19．67±4．53）％、（39±17．4）％，差别显著。实验Ⅰ组与空白组相比无显著性差别。结论丹参注射液预防顺铂耳毒性作用与给药时间有关；认为丹参在顺铂使用后6h给药其预防顺铂耳毒性的作用更为显著。Objective To explore the effects of Administration time of Salvia miltiorrhiza injection on precaution the ototoxicity of Cisplatin.Methods 40 guinea pigs were randomly divided into 4 groups： control group, experiment group Ⅰ, experiment group Ⅱ and empty group. Control group were injected Cisplatin 1 mg/kg/d by intraperitoneal. Eexperiment group Ⅰ ,6 hours after injection ofCisplatin1mg/kg/d by intrapefitoneal,were injected Salvia miltiorrhiza injection 8mL/kg/d by intraperitoneal. Experiment group Ⅱ were injected Cisplatin 1 mg/kg/d and Salvia miltiorrhiza injection 8mL/kg/d by intraperitoneal simultaneously.And empty group were injected normal sodium 0.8mL/kg/d by intraperitoneal. One time everyday, 6 days in all. Before and after the injection,BEAP was detected,and reaction threshold of ABR Ⅲ was taken as detected audibility threshold（dB）.Results Audibility thresholds from control group,experiment group Ⅰ and experiment group Ⅱ were all rised,and they were （42.6 ± 4.32）dB,（33.0 ± 3.00） dB and （36.6 ± 2.16）dB respectively.Audibility thresholds from empty group was （31.5 ± 2.25）dB.Experiment group Ⅰ and experiment group Ⅱ were lower than control group,P〈0.05. The augmented rate： experiment group Ⅰ 〈 experiment group Ⅱ 〈 control group, ang they were （6 ±8.4）%,（19.67 ±4.53）% and （39 ± 17.4）%, P〈 0.05.There was no significant defference between experiment group Ⅰ and empty group.Conclusion It is related with administration time that the effect of Salvia miltiorrhiza injection on precaution the ototoxicity of Cisplatin;and 6 hours after Cisplatin using Salvia miltiorrhiza injection, the effect is more significant.
