摘 要:目的:利用超声白蛋白微泡造影剂经一定声压超声辐照后产生的"声孔效应"(Sonoporation),以及其靶向性来提高前列腺内的药物浓度。方法:选取已建立慢性前列腺炎模型大鼠30只,随机分为对照组,给药组和给药加微泡组。将EA-10原料药以溶媒溶解后注射给药。对照组给予生理盐水,给药组以15mg/kg剂量给药,微泡组以15mg/kg剂量给药,给药后微泡尾静脉注射并用超声基因转染仪前列腺局部间歇辐照2min。照射后取前列腺组织,匀浆后测量组织中EA-10的浓度变化。结果:对照组前列腺组织中没有药物,给药组和给药加微泡组前列腺组织中药物浓度较对照组高(P<0.01),给药加微泡组前列腺组织中药物浓度较给药组高(P<0.05)。结论:超声白蛋白微泡造影剂的"声孔效应"对于前列腺组织药物浓度的提高有明显的促进作用。Objective:To increase the medicine concentration in rat prostate by the "sound aperture effect" and site-targeted effect of ultrasound-Mediated Microbubble in order to get better therapeutic effect. Methods: 30 1-year-old male Sprague-Dawley rats were castrated and injected estradiol benzoate for 30 days and divided into three groups randomly, model group, treatment group and ultrasound-mediated microbubble group. Model group was treated with sodium chloride, treatment group was treated with EA-10 and ultrasound-mediated microbubble group was given ultrasound-mediated microbubble and EA-10. Concentration of EA-10 in prostate was measured after the prostate was taken and homogenated. Results: Concentration of EA-10 in treatment group as higher than that of in model group(P〈0.01). But more EA-10 was measured in ultrasound-mediated microbubble group than in treatment group(P〈0.05). Conclusion: Ultrasound-mediated microbubble can markedly promote the increase in the drug concentration in rat prostate.
